Directions: Metagenics Bio Absorb PEA Advanced
Adults: Take 1 capsule twice daily or as directed by your healthcare professional.
Dosing – The daily dosage of PEA in clinical trials has ranged from 300mg–1200mg daily for chronic pain and up to 1800mg daily for acute colds and flu. Clinical trial data would suggest that a loading dose of 600–1200mg for 3–4 weeks followed by a maintenance dose of 300–600mg may be the most appropriate dosing strategy for chronic pain conditions. Individual requirements will vary.
Timing – Considering that PEA is a fat soluble compound, consumption with a fat-containing meal may enhance absorption.
Contraindications and adverse effects – There are no known contraindications. PEA has been clinically studied across a broad population group and has been found to be highly tolerable, with aside effect profile similar to placebo.
Why is there a 21-Day Warning for Bio Absorb PEA? Is it safe to take long term?
The Therapeutic Goods Association (TGA) mandates a 21-day warning be placed on all listed PEA product labels. In listing this medicine, 17 clinical studies were provided to the TGA, of these, only three were conducted in ‘healthy participants’ with the maximum duration of these studies being 21 days, hence these three studies were considered by the TGA.
Numerous other studies exist which used PEA over three months without negative effects. See references Tab for 256 references relating to PEA.

Palmitoylethanolamide (PEA) is neuroprotective, anti-inflammatory, and analgesic4 due to its endocannabinoid (eCB) action.5 PEA affects eCB signalling through peroxisome proliferator-activated receptor alpha (PPAR-α) activation6and is capable of exerting antiallodynic and anti-hyperalgesia effects by downmodulating both microglial and mast cell activity which has led to its use for neuropathic pain.7Both TRPA1 (transient receptor potential ankyrin 1) and TRPV1 (transient receptor potential vanilloid type 1) are co-expressed in sensory neurons and are implicated in neuropathic pain, hyperalgesia and neural amplification.8 Animal studies have shown that a main phytoconstitutent of saffron, safranal, desensitises TRPA1 and PEA desensitises TRPV1.9 In addition, vitamin B1 deficiency results in neuropathic pain10 and vitamin B1 was found to significantly reduce dorsal root ganglia neuron excitability in vitro, a provocating factor in central sensitisation of pain.11

Levagen+ uses LipiSperse technology for enhanced absorption24 and is 70% better absorbed (Figure 2) leading to greater plasma levels over time compared to a standard PEA. 600 mg/d of Levagen+ PEA is likely to equate to consuming 1,000 mg of a standard micronized PEA.
