How increasing NAD+  promotes healthy ageing

Nicotinamide adenine dinucleotide (NAD+) is an essential molecule that plays a critical role in almost every metabolic process in the body.

Through its roles as a cofactor, electron donor and signalling molecule, NAD+ is essential for many biological processes. These include ATP synthesis, DNA repair, secondary messenger signalling, epigenetic regulation of gene expression and cell survival.

NAD+ naturally declines with age. The gradual decline of NAD+ concentrations takes place across multiple human organs and tissues, including skin, blood, liver, muscles and brain. Reduced concentration of NAD+ results in suboptimal functioning of the NAD+ dependent pathways. This in turn results in an increase in issues relating to metabolic function, inflammation, senescence and degeneration of the nervous system. When NAD+ dependent mechanism are compromised, diseases of ageing are then more likely to occur.

What is nicotinamide mononucleotide (NMN)?

NMN is a naturally occurring molecule that helps produce energy in the body.

NMN is a molecule called a nucleotide. Nucleotides have many roles in the body including the production of DNA.

NMN is converted into another molecule within the cells known as nicotinamide adenine dinucleotide (NAD). Your body needs NAD for a variety of functions involved in metabolism and energy production.

The amount of NAD your body can make depends on the amount of NMN available in your body.

What is the difference between NR, NMN and NAD?

In the body, NR (Nicotinamide Riboside) converts to NMN (Nicotinamide Mono Nucleotide) which is then converted in the cell to NAD (Nicotinamide Adenine Dinucleotide).  

The best form to take is NMN (Nicotinamide Mono Nucleotide).

Is it better to take a supplement containing NMN or NAD?

Research has found that taking a supplement containing NMN is better than taking a NAD supplement. This is because NAD is unable to enter the cell because the cells have receptors for NMN but do not have receptors for NAD. So, it is best to take NMN because NMN can enter the cells where it is converted to NAD.

Researchers who study NMN and NAD believe that the goal is to get NAD levels in the cells to levels closer to those levels in our youth.

Studies in mice have found that the gut absorbs NMN readily. More studies are needed to investigate how eating foods containing NMN may increase NAD levels in cells.

Can NMN  help with ageing?

NMN and NAD levels naturally decline with age. Researchers have suggested that taking NMN supplements may help with age-related health concerns.

A study in 10 Japanese men found that taking NMN at 100, 250, or 500mg increased levels of NAD. 

Insulin resistance can develop as we age and is one of the reasons diabetes is more common in older adults. Although studies haven’t specifically looked at the effects of taking NMN supplements, the research available suggests that it may help you maintain or regain insulin sensitivity with age.

More studies on these effects in humans are needed to investigate this potential benefit
